Start up

  1. Remove the dust cover and put it on the shelf above the microscope. 
  2. If you are taking brightfield images, turn on the switch at the bottom left of the microscope stand.
  3. If you are imaging in fluorescence, switch on the Sola Light Engine and press the Light button.
  4. Switch on the power for the extension lead on the wall.  Check that the switch on the extension lead itself is also on.  This will power the camera and the computer.
  5. Switch on the computer (on the floor under the table).
  6. Log in to the LAS-X user account.  The password is lmcb.
  7. Start the LAS X software.

LAS X

  1. To acquire images,  select select Acquire and the ACQUIRE Acquisition tab and "Acquisition".Image Removed.

  2. You can save the correct objective into your image metadata by selecting the magnification you are using (under magnificationMagnification).  As the turret is manually operated the   As the turret is manually operated the objectives are changed by hand.  There is also a 1is also a 1.5x optovar that can be put magnification changer that can be put into the light path using the wheel on the right of the microscope stand and stand and changing the "magnification changerMagnification Changer" drop down selection.  Make sure to let the software know knows whether the 1.0x or 1.5x optovar lens is in place, otherwise your scales image scaling will be offwrong.
  3. To see a live view image,  click click 'liveLive';  to to take an image,  click click 'capture imageCapture Image'. 
  4. Select colour or black and white mode by using the selectors.  Note : We advise using black and white mode for all fluoresecently stained specimens, and merging them later in post processing.  Image Removed

  5. For brightfield images, set the white balance using the "draw rectangle" tool and selecting 'white balance' when the options appear.
  6. Set the exposure time in the imaging tab.  Leave the Gain at 1.
  7. To save, go to 'Open projects', right click on your file or folder, and save.  LAS X will save whole folders of images into one single lif.  If you want individual tifs, use the right click > export option.
