Start up

  1. Remove the dust cover and put it on the shelf above the microscope.  DO NOT PUT THE DUST COVER ON THE FLOOR.
  2. If you are taking brightfield images, turn on the switch at the bottom left of the microscope stand.
  3. If you are imaging in fluorescence, switch on the ebq100 power supply to the mercury lamp.  Please refer to the LMCB Light Microscopy Local Rules for the Mercury Lamp Safety Rules. 
  4. Switch on the power for the extension lead on the wall.  Check that the switch on the extension lead itself is also on.  This will turn power on to the camera and the computer.
  5. Switch on the computer (on the floor under the table).
  6. Log in to the LAS-X user account.  There is no password.
  7. Start the LAS X software.






LAS X

  1. To acquire images, ensure that you have selected the ACQUIRE tab and in "Acquisition".

  2. You can save the correct objective into your image's metadata by selecting the magnification you are using (under magnification).  However as it is a manual turret, you'll have to change the objectives by hand.  There is an additional 1.5x optovar you can put into the light path using the wheel on the right of the microscope stand.  Let the software know by changing the "magnification changer" drop down selection.
  3. To see a live view of your image, use the 'live' button, and to take an image, use the 'capture image' button. 
  4. Select colour or black and white mode by using the selectors.  Note : We advise using black and white mode for all fluoresecently stained specimens, and merging them later in post processing. 
  5. For brightfield images, set the white balance by using the "draw rectangle" tool and selecting 'white balance' when the options appear.
  6. Set the exposure time in the imaging tab.  Leave the Gain at 1.
  7. To save, go to 'Open projects', right click on your file or folder, and save.  LAS X will save whole folders of images into one single lif.  If you want individual tifs, use the right click > export option. 

Shutting down

  1. Shut down windows

  2. Turn off the ebq100 mercury lamp
  3. Turn off the wall switch labeled 'extension lead'
  4. Put the dust cover back onto the microscope
