Advantages of this microscope
Use this microscope for specimens that can only be imaged in open chambers in aqueous media (e.g. tissue, pupae/embryos, fish) or that require a longer working distance (the water dipping lenses have working distances in the order of 2 mm). Live mammalian cells can be imaged if the imaging chamber can be inverted so the cells are upside down. There is a heated stage adapter for warming the specimen but there is no CO2 supply so the chamber must be sealed to prevent pH change. This microscope can also be used to image slides. There is no 405 nm laser, so you cannot image DAPI or Hoechst.

458 | Argon | CFP | This is not a very powerful laser line | 476 | Argon | - | Could be used as an alternative for green dyes when trying to separate dyes with similar spectra | 488 | Argon | EGFP, FITC, Alexa 488 |
| 514 | Argon | YFP | Could also be used for Citrine, Venus, etc | 543 | GreenHeNe | TRITC, Alexa 532, Alexa 546, Alexa 555, Cy3 | Can also be used for 'redder' dyes like Texas Red, Alexa 568, Alexa 594, mCherry but with lower excitation efficiency | 637 | Red Diode | Cy5, Alexa 633, Alexa 647, Si Rhodamine dyes, DRAQ5 |
|
Note that there is no 405 nm laser on this machine so you cannot image Hoechst, DAPI or anything else in the blue region of the visible spectrum Links |
