Warner Instruments perfusion equipment


The LMCB light microscopy facility has several items of equipment from Warner Instruments for perfusion and environmental control of chambers inserted into 35 mm dishes. Warner's equipment and consumables is very flexible and can be set up in many ways to make many kinds of perfusion systems. Unfortunately this also makes it rather tricky and time-consuming to work out how to put it all together. The simple guides below will hopefully provide enough information to allow you to assemble equipment for use in microscope environmental chambers and other situations. There are also manuals and technical notes from Warner Instruments that are stored with the equipment itself. Warner Instruments RC-37F - perfusion apparatus with two syringes

The image above shows a Warner Instruments RC-37F insert for Falcon 35 mm dishes mounted in a Mat-Tek glass-bottomed dish. The glass-bottom dish is mounted in an MDA-1 mini-dish adapter for Warner RC-20 series chambers. The MDA-1 clamps are being used to hold the RC-37F in the dish. The MDA-1 is further mounted in an SA-20-MW-AL stage adapter for multi-wells. This will fit in any microscope specimen adapter designed for multi-well plates.

The RC-37F inlet and outlet ports are connected to short lengths of TY-50 Tygon 1/10” tubing which are in turn connected by TC-3 tubing connectors to SL-6 Luer stopcocks and standard plastic syringes. The TY-50 tubing can be cut to any length.

A. This image illustrates the overall appearance of a syringe manifold for supplying fluid to the RC-37F insert for 35 mm dishes. The flow is controlled using a pinch valve assembly that is controlled by switches on a VC-6 six channel valve controller.

B. A 50 ml syringe is mounted in the syringe manifold and connected to TY-50 Tygon 1/10” tubing by an SL6 Luer stopcock connected to a tubing adapter (TC-3)

C. A short piece of C-FLEX tubing is inserted into one of the pinch valves. This C-FLEX tubing is connected above and below the valve to lengths of TY-50 Tygon 1/10” tubing with tubing adapters (TC-3).

D. The RC-37F insert is mounted in a 35 mm Mat-Tek glass-bottomed dish. The dish is mounted in an MDA-1 mini-dish adapter for the RC-20 series of Warner chambers. The MDA-1 clamps are used to hold the RC-37F in the dish. The MDA-1 is further mounted in an SA-20-MW-AL stage adapter for multi-wells, which will fit in any microscope specimen adapter designed for multi-well plates. The RC-37F inlet port is connected to an MP-2 manifold using short pieces of PE-160 polyethylene tubing. The outlet port is attached to a short piece of C-FLEX tubing. Warner Instruments recommend attaching the outlet to a vacuum line, but a peristaltic or syringe pump might also work.
