The SPE3 is a 2012 model Leica TCS SPE confocal microscope mounted on a semi-automated, inverted Leica DMI4000 stand. The stand is capable of fluorescence, bright-field and DIC techniques and is fitted with a motorised stage. The confocal system has four lasers and a single photomultiplier tube (PMT) detector so multichannel acquisition must be done sequentially. The microscope stand has a manual focus drive so optical sectioning is achieved using the Z Galvo insert integrated into the stage. The software has Tile Scan and Mark & Find modes, which use the microscope's motorised stage to revisit saved positions or build up a 'mosaic' of images of adjacent areas.
