Leica TCS SP5 Confocal Microscope

Location: MRC Building, first floor, room 1.20B

The SP5 is a flexible, high-end laser scanning confocal microscope suitable for fixed and live samples. It is built around an inverted DMI6000B microscope capable of fluorescence, bright-field and DIC techniques. The SP5 is a tandem scanner system, which means that it has both a conventional scanner and a fast (8000Hz) resonant scanner for high speed imaging. It is fitted with 5 lasers, producing 8 potential excitation lines, and has 4 PMT detectors and one transmitted light detector. Optical sectioning of samples is achieved using the microscope's integrated focus drive. The software has Tile Scan and Mark & Find modes, which use the microscope's motorised stage to revisit saved positions or build up a 'mosaic' of images of adjacent areas. FRAP, FRET AB, FRET SE and Live Data Mode modules are also included with the software. A Pe-Con stage-top incubation system with heat and CO2 control is available for live mammalian samples.
